About this test
When a sore throat refuses to settle or comes with high fever, this test answers the question a quick look in the mirror cannot: is a treatable bacterium behind it, and which antibiotic will actually clear it? A cotton swab is rubbed firmly over the back of the throat and tonsils to collect any organisms present, and the sample is then streaked onto culture plates and incubated for two to three days. The laboratory watches for growth of significant pathogens, the most important being Group A beta-haemolytic Streptococcus (Streptococcus pyogenes) โ the cause of classic strep throat โ but the culture also picks up other relevant bacteria such as Staphylococcus aureus, Haemophilus, and occasionally diphtheria-causing organisms or fungal overgrowth. The accompanying sensitivity (the C/S part) tests how each isolated bacterium responds to a panel of antibiotics, so your doctor prescribes a drug that works rather than guessing. Doctors typically order it for a painful throat with fever, swollen tender neck glands, white patches or pus on the tonsils, difficulty swallowing, and absence of a runny nose or cough that would point instead to a simple virus. Confirming and properly treating Group A Streptococcus matters well beyond the sore throat itself: untreated infections can lead to rheumatic fever and rheumatic heart disease, which remain meaningfully common in Nepal among children and young adults. Results are read together with your symptoms, a throat examination, and sometimes a rapid strep test or blood count, and the sensitivity report guides antibiotic choice especially where resistance or penicillin allergy is a concern.

Preparation
No fasting needed. Come before starting any antibiotic if possible, as even one dose can stop the bacteria growing. Avoid antiseptic mouthwash, gargles, or lozenges on the morning of the test, and do not eat or brush your teeth in the hour beforehand. The swab takes only a few seconds and may briefly make you gag.
